摘要目的研究大孔树脂分离纯化甘草总黄酮的最佳树脂和最有工艺为甘草总黄酮的工业化生产提供实验依据。方法:分别对DS401、D101、AB8、HPD100四种大孔树脂进行静态吸附、静态解吸实验筛选用于甘草总黄酮分离的最佳树脂,采用紫外分光光度法测定总黄酮质量浓度,以总黄酮吸附率、洗脱率、收率、纯度等指标评价AB8大孔树脂分离纯化甘草总黄酮工艺种上样量、上样液质量浓度、吸附流速、洗脱剂乙醇体积分数极其用量对吸附和解吸效果的影响,从而确定最佳工艺。结果:AB8型大孔树脂为分离甘草黄酮类组分最佳树脂,其分离的最佳工艺为:总黄酮浓度为920mgmL1的样液,吸附流速为2mlmi
1洗脱剂50乙醇用量为4倍树脂体积,利用该工艺精制后的干草总黄酮纯度和收率分别为7445、3531。结论:获得的样品总黄酮纯度达70以上,但收率较低,综合各方方面因素,该工艺简单可。关键字甘草;总黄酮;分离纯化;大孔树脂
